Abstract

Objective: To document the first official isolation of Hirsutella citriformis Speare in Colombia, describing their morphological, morphometric, and molecular characterization, as well as its pathogenicity against Diaphorina citri, vector of Huanglongbing (HLB) in citrus. Scope: The research aims to position H. citriformis as a viable biological alternative for managing D. citri, contributing to the sustainability of the Colombian citrus industry. Methodology: Infected psyllid insects with H. citriformis were collected at the Montelindo Farm of the University of Caldas. The strain was isolated, purified, and characterized using morphological, morphometric, and molecular techniques. A bioassay was conducted to evaluate their pathogenicity through direct contact between the insects and sporulated cultures. Main results: The strain exhibited morphological and morphometric characteristics typical of H. citriformis, with reproductive structures within previously reported ranges (phialides with bulbous base, long and thin neck ending in an elongated conidium, covered with a mucilaginous layer). Sequence analysis using the 28S ribosomal gene the identification of the species as Hirsutella citriformis was confirmed with 98,86% similarity. The accumulated mortality in the D. citri bioassay reached 98% in the treatment with H. citriformis conidia, and 3% in the control. Treated insects developed synnemas 7 days post-mortem. Conclusion: According to the morphological, morphometric, and molecular characteristics of the isolates, the strain was identified as belonging to the species H. citriformis Speare in addition, was shown to be pathogenic for against D. citri.
